Human induced pluripotent stem cells and male infertility: an overview of current progress and perspectives

چکیده
Recently, significant progress has been made in ART for the treatment of male infertility. However, current ART has failed to help infertile patients with non-obstructive azoospermia, unless donor sperm is used. In fact, most couples wish to have their own genetically related child. Human induced pluripotent stem cells (hiPSCs) can be generated from patients' somatic cells and in vitro derivation of functional germ cells from patient-specific iPSCs may provide new therapeutic strategies for infertile couples. The overall developmental dynamics of human primordial germ cells are similar to that in mice, but accumulating evidence suggests that there are crucial differences between human and mouse PGC specification. Unlike mouse iPSCs (miPSCs) in naive state, hiPSCs exhibit a primed pluripotency which possess less potential for the germ cell fate. Based on research in mice, male germ cells at different stages have been derived from hiPSCs with different protocols, including spontaneous differentiation, overexpression of germ cell regulators, addition of cytokines, co-culture with gonadal cells in vitro and xeno-transplantation. The aim of this review is to summarize the current advances in derivation of male germ cells from hiPSCs and raise the perspectives of hiPSCs in medical application for male infertility, as well as in basic research for male germ cell development.

Background: We have recently shown that human embryonic (hESCs) and induced pluripotent stem cells (hiPSCs) can differentiate into advanced spermatogenic cells including round spermatids by in vitro culture (Easley et al., Direct differentiation of human pluripotent stem cells into haploid spermatogenic cells. Cell Reports 2, 440-446 2012) and also, in collaboration, that rhesus spermatogonial ...

Successful isolation, derivation and culturing of human pluripotent stem cells, including human embryonic stem cells (hESCs) and human induced pluripotent stem (hiPSCs) cells in laboratory scale has opened new horizones for cell therapy applications such as tissue engineering and regenerative medicine. However, most of the cell therapy protocols using these unique cells require large number of ...
متن کاملA Quick update from the Past to Current Status of Human Pluripotent Stem Cell-derived Hepatocyte culture systems
Pluripotent stem cells (PSCs) may be offered as an unlimited cell source for the hepatocyte generation. The generation of hepatocytes from stem cells in vitro would provide an alternative cell source for applications in drug discovery and cell transplantation. In this review, we discuss different approaches to generate pluripotent stem cell-derived hepatocytes, advantages, limitations for each ...

Stem cells are undifferentiated and multi pluripotent cells which can differentiate into a variety of mature cells and tissues such as nervous tissue, muscle tissue, epithelial tissue, skeletal tissue and etc. Stem cells from all different source have three unique features: 1) Proliferative capability: Stem cells are capable of self dividing and self renewing for long periods or more than six m...
متن کاملStem Cells as New Agents for the Treatment of Infertility: Current and Future Perspectives and Challenges
Stem cells are undifferentiated cells that are present in the embryonic, fetal, and adult stages of life and give rise to differentiated cells that make up the building blocks of tissue and organs. Due to their unlimited source and high differentiation potential, stem cells are considered as potentially new therapeutic agents for the treatment of infertility. Stem cells could be stimulated in v...
